Renai and Josh Lambert have recently moved into a new house with their three children. One morning, Renai begins looking through a family photo album with her son, Dalton. He asks why there are no pictures of Josh when he was a child. Renai reasons that he has always been camera shy. Dalton tells Renai he is scared of his new room; she calms him down and stacks a few books on a nearby shelf. When she finds the books lying on the floor later, Renai tells Dalton to put them back after he looks at them. Dalton claims that he did not touch them. Sometime later, as Dalton is playing, he hears noises in the attic and goes to investigate. He steps on an old ladder to turn on the light, when it breaks, causing Dalton to fall. He starts screaming after looking at something offscreen in a shadowy corner. Renai and Josh help him, then tuck him into bed, confident he only took a minor fall and scarred himself. The next day Josh goes to wake Dalton, but he does not move. They rush him to the hospital where the doctors say he is in an unexplained coma. Three months later he is still in a coma and they bring him home to care for him.

11-year-old Greg Heffley is anxious to start middle school, confident he will easily become the school's most popular kid. However, Greg worries about how his "uncool and embarrassing" best friend, Rowley Jefferson, will fit in. While Rowley is a good friend who helps Greg escape his terrorizing older brother, Rodrick, Greg worries that Rowley's "dorky" clothes and personality will embarrass both of them.

On their first day, Greg and Rowley meet Angie Steadman, a seventh grader who works for the school's paper. Throughout the school year, Angie gives Greg and Rowley information about the workings of middle school, which she believes was created to store kids while they make the awkward transition between children to teenagers. Classmate Chirag Gupta tells Greg and Rowley the story behind the moldy slice of Swiss cheese laying on the school's basketball court; upon touching the cheese, a kid named Darren Walsh contracted the Cheese Touch; anyone who touches the cheese will create chaos as they pass on the Cheese Touch by touching someone else. The Cheese Touch eventually got taken away by a German exchange student named Dieter Müller, who moved back to Düsseldorf and took the Cheese Touch with him.

Greg and his family attending a skating rink for the year's new seventh graders. Here he reunites with his best friend Rowley and classmates Fregley and Chirag. Greg then meets Holly Hills, who immediately becomes his love interest. While attempting to skate with Holly, Greg's older brother Rodrick intervenes, ending with their mother speaking to him on the loudspeaker and further humiliating him by getting Greg's father to carry him off the rink.

As weeks go by, Greg's mom notices that the boys do not get along, and calls for a family meeting. She gives them a new incentive to get along together by rewarding them with "Mom Bucks" whenever they spend time together, which they can trade in for real money. The boys eagerly accept and compete with each other to earn the Mom Bucks.

After a talent show is advertised on TV, the brothers see this as a big opportunity--Rowley suggests that he and Greg perform magic tricks, whilst Rodrick sees this as his band's big break. Greg films Rowley lip-synching "Tik Tok" by Ke$ha and uploads it to YouTube, hoping that it will become an online hit, but much to his dismay, it only receives four views.

On Sunday, the Heffley family goes to church. Greg is extremely reluctant to go inside, as a melted 3 Musketeers bar, intentionally placed on his seat by Rodrick, has stained his trousers. Greg's mom uses her blouse to cover up the stain, but the cover-up backfires when the family go to take communion and Rodrick exposes Greg's pants. The boys ultimately end up in a scuffle on the floor.

Melissa Morris (Emily Osment) desperately tries to gain more attention from her always preoccupied father, Neal, a best-selling author (George Newbern). Melissa lives in the shadow of Tripp Zoome (Jonathan Keltz), the adventurous and clever hero from her father's popular spy novels. Before a long overdue father-daughter vacation, her father gets kidnapped by obsessive fans: Wheeze (David Henrie) with his friend, Sheldon (Denzel Whitaker), and his brother, Andre (Moises Arias).

The tables turn however, when she must team up with them to find and rescue him from Merv (Jason Earles), and brothers Maurice (Phill Lewis) and Skunk (Charles Halford). It turns out Merv is just using Skunk and Maurice as part of a plan to get Neil to write one final book using his daughter as a reader.
